"Procedures to Assess the Hazard of a Superfund Site"
The major aims of this project include (i) synthesis of analytical and toxicological standards; (ii) development of in vitro bioassays (P4501A1 induction, antiestrogenicity, DRE binding); (iii) in vivo bioassays to determine TEFs for polynuclear and halogenated aromatic hydrocarbons; and (iv) application of TEFs for site extract risk assessment.

"Effects of 2,3,7,8-TCDD on Estrogenic Responses"
This project is focused on the molecular biology of TCDD-induced antiestrogenicity in MCF-7 cells and utilizes 5'-regulatory sequences from diverse estrogen-regulated genes to identify cis-acting genomic sequences and trans-acting factors. The role of DRE and DRE-Sp1 motifs as enhancer sequences in the cathepsin D gene will also be investigated.

This is a toxicology training grant proposal supporting six graduate research assistants and one postdoctoral fellow. S. Safe is the PI on this proposal, and there are nine co-PIs. Support is for two graduate assistantships.

"Inhibitory Ah Receptor-Androgen Receptor Crosstalk in Prostate Cancer"
This project will investigate the mechanisms of inhibitory AhR-AR interactions in prostate cancer and will determine if inhibitory AhR-AR crosstalk blocks androgen-induced cell cycle progression.

"Mechanisms of Growth Factor-Estrogen Receptor Crosstalk"
The major aims of this project are to define molecular mechanisms of growth factor-estrogen receptor crosstalk associated with induction of cathepsin D and c-fos expression in breast cancer cells. The mechanism of inhibition of growth factor-induced responses by TCDD will also be investigated.
